摘要
目的测定金荞麦中金丝桃苷的含量。方法采用HPLC法,色谱柱为Shimadzu C18(150mm×4.6mm,5μm),流动相为甲醇-0.2%H3PO4溶液(45:55),流速为1.0ml·min-1,检测波长为350nm,柱温为室温,用标准曲线法定量,测定金荞麦中金丝桃苷的含量。结果金丝桃苷0.5~2.5μg与峰面积的线性关系良好(r=0.9998),回收率为95.01%,RSD=2.94%。结论所建方法简便、快速,适用于金荞麦中金丝桃苷的测定。
OBJECTIVE To determine the content of hyperoside in Fagopyrum dibotrys. METHODS An HPLC method was used. The analysis was carried out on Shimadzu C18 column( 150 mm×4. 6 mm,5μm). The mobile phase was M eOH- 0. 2% H3 PO4 (45 : 55 ) with flow - rate of 1.0 ml.min^-1. The eluate was detected at 350 nm in room temperature. RESULTS The method had a good linear relationship. The linear range of hyperoside was 0. 5 - 2.5 μg( r = 0. 9998). The recovery was 95.01% with RSD of 2. 94%. CONCLUSION The method is simple and suitable for the determination content of hyperoside in Fagopyrum dibotrys.
