茉莉直接不定芽途径高效再生体系的建立 被引量:3

Establishment of Efficient Regeneration System of Direct Adventitious Shoot in Jasminun Sambac

摘要 以茉莉的下胚轴为外植体,研究了茉莉直接不定芽器官发生途径的再生体系.结果表明:以MS为基本培养基,附加0.5 mg/L BAP和0.01 mg/LNAA的诱导效果最佳,其不定芽再生率高达92%,外植体平均不定芽数目为4.2.适宜不定芽增殖的最佳培养体系为MS+1.0 mg/L BAP+0.1 mg/L NAA.1/2MS+0.5 mg/L IBA+0.5 mg/LIAA+100 mg/L适于再生幼茎的生根,生根率达91%,生根后经移栽成活. Hypocotyls of Jasminun sambac was used as explants, and the regeneration system of highly efficient direct adventitious shoot on Jasminun sambac were studied. The results showed that the best induction was MS medium supplemented with 0.5 mg/L BAP and 0.01 mg/L NAA. Ninety - two percent of regeneration frequency and 4.2 shoots per explants were obtained after 30 days of culture. Regenerated shoots proliferated efficiently on a shoot multiplication medium consisting of MS medium containing 1.0 mg/L BAP and 0.1 mg/L NAA. Regeneration tender stems were rooted on a rooting medium made up of MS medium enriched with 0.5 mg/L IBA and 0.5 mg/L IAA. After hardening, 90% of plants were successfully established under greenhouse donditions.
