江西3种红鲤血清转铁蛋白遗传多态性 被引量:3

Serum Transferrin Genetic Polymorphism of Three Breeds of Red Common Carp in Jiangxi Province

摘要 采用不连续聚丙烯酰胺垂直电泳(PAGE)和不连续十二烷基磺酸钠-聚丙烯酰胺垂直电泳(SDS-PAGE)对兴国红鲤、玻璃红鲤、荷包红鲤的血清转铁蛋白(Tf)进行研究。PAGE电泳结果表明3种红鲤的Tf图谱具有3条带,荷包红鲤与玻璃红鲤Tf表型由Tfb、Tfc组成,兴国红鲤Tf表型由Tfa、Tfb、Tfc组成;其中玻璃红鲤的Tfb和Tf c条带表达量最高,荷包红鲤的Tfb、Tfc条带表达量最低。不连续SDS-PAGE电泳结果显示3种红鲤Tf的分子量相近,为68 k Da^80 k Da。结果表明转铁蛋白可用于3种红鲤亲缘关系的分析,它们的表达存在一定差异,但三种红鲤鱼的亲缘关系较近。 Serum transferrin (Tf) polymorphism of three breeds of red common carp in Jiangxi Province, namely C. carpio var. Wuyuannensis, C. carpio var. Xingguonensis and C. carpio var. Wanannensis, were analyzed by native polyacrylamide gel electrophoresis (PAGE) , and the molecular weight of Tf was determined by sodium dodecylsulfate polyacrylamide gel electrophoresis ( SDS - PAGE). There were three Tf bands in the three breeds of red common carp. The phenotype of the C. carpio var. Wuyuanensi and C. carpio var. Wananensis Tf was composed of Tf b and Tfc bands,the phenotype of the C. carpio var. Xinguoensis Tf was composed of Tf a,Tf b and Tf c bands;The concentration of Tfb ,Tfc bands in the C. carpio var. Wananensis was the highest and the concentration of Tfb ,Tfc bands in C. carpio var. Wuyuanensi was the lowest among them. The result of SDS - PAGE showed that the molecular weights of three breeds of red common carp were similar different and from 68 kDa to 80 kDa. The results might suggest that serum transferrin can be used for analysis of genetic relationship among three breeds of red common red carp for their different expression, and it also indicated their genetic relationship was closer.
