摘要
用超临界CO2流体萃取技术从曼地亚红豆杉枝条中提取紫杉醇。在分离釜Ⅰ和Ⅱ温度为30℃、压力为6.5MPa、CO2体积流量为25L/h条件下,正交实验优化所得萃取过程的最佳工艺参数为:萃取时间3.5h;萃取压力30MPa;萃取温度47℃;夹带剂为φ(CH3CH2OH)=85的乙醇溶液。萃取方式为静态和动态萃取相结合。最佳工艺条件下各用35.0g原料进行3次验证实验,紫杉醇的平均提取量为9.58mg,平均提取率87.76;浸膏中紫杉醇质量分数为1.12,高于传统的溶剂浸提(浸膏中紫杉醇的质量分数约0.87)。
Supercritical CO2 fluid extraction technology was used to extract taxol from the branches of Taxus media. With the preconditions of separation temperature 30℃, pressure 6. 5 MPa and CO2 flux 25 L/h, the best process conditions were extraction time 3.5 h, pressure 30 MPa, temperature 47℃, the entrainer φ(CH3CH2OH)=85 % , and the manner of conbined static and dynamic extraction. Under the optimal conditions, validatory experiments were carried out for three times with the average extraction mass of taxol 9.58 mg and extraction rates 87.76%. In these crude extracts, the average content of taxol was 1.12% ,which was higher than that of traditional solvent extraction(the content of taxol was about 0.87%)
