摘要
为建立鸡蛋中三聚氰胺残留的简便高效液相色谱法,本试验采用NH2柱作为色谱分析柱,90%乙腈水溶液为流动相,鸡蛋样品经混合均匀后,用三氯乙酸-乙腈溶液提取,经阳离子交换固相萃取柱净化、富集后,在紫外检测器230nm处用高效液相色谱仪测定,保留时间在6~7min。标准曲线在0.1~5.0μg/ml浓度内呈线性相关,相关系数(r)为0.9999。在空白鸡蛋样品中添加0.2mg/kg、0.5mg/kg、2.0mg/kg和5.0mg/kg的三聚氰胺,回收率为78.96%~92.19%,相对标准偏差(RSD)为1.96%~6.60%。样品检测定量限为0.2mng/kg。
A high-performance liquid chromatography (HPLC) with NH2 column and 90% acetonitrile eluate to determine the melamine in eggs was developed. Samples were extracted with 1% trichloroacetic acid-acetonitrile. Target analyres of samples were cleaned up and concentrated by HyperSep Retain-CX column solid-phase extraction (SPE). A perfect separation for melamine was achieved and retention time was about 6 - 7 min at 230 nm. The calibration curve was linear in a concentration range of 0. 1 - 5.0 g/ml, with correlation coefficient of 0. 999 9. Recoveries of melamine at fortified levels of 0. 2 mg/kg, 0. 5 mg/kg, 2. 0 mg/kg and 5.0 mg/kg ranged from 78.96% -92. 19%, with relative standard deviation of 1.96% - 6. 60%. The quantification limit was 0. 2 mg/kg.
