摘要
目的:观察乙酰半胱氨酸及扎鲁斯特对急性肺损伤的防治作用。方法:大耳白兔32只,随机分为对照组、致伤组、乙酰半胱氨酸治疗组和扎鲁司特治疗组,每组8只。致伤组经颈静脉注入脂多糖(LPS,1 ml/kg);对照组注入生理盐水(1 ml/kg);乙酰半胱氨酸治疗组在注入LPS(1 ml/kg)前2 min,经颈静脉注入乙酰半胱氨酸(300 mg/kg);扎鲁斯特治疗组在注入LPS(1 ml/kg)前1 h及0.5 h,胃内注入扎鲁司特(20 mg/kg)。测定各组肺动脉压、动脉血氧分压、外周血白细胞计数、肺泡灌洗液(BALF)NO2-浓度、肺系数(肺湿重/体重)及肺湿/干比。结果:乙酰半胱氨酸组肺动脉压及NO2-浓度与致伤组有显著差别(P<0.05),乙酰半胱氨酸及扎鲁斯特组氧分压均较致伤组明显改善(P<0.05),肺系数和肺湿/干比显著均低于致伤组(P<0.05),外周血白细胞高于致伤组(P<0.05)。结论:乙酰半胱氨酸和扎鲁斯特对兔内毒素急性肺损伤有一定的保护作用。
Objective:To observe the preventive and cure effects of N -acetyl - L - cysteine and zafirlukast on acute lung injury. Methods:32 rabbits were randomly divided into 4 groups (n = 8) : control group (CG) , injury group (IG) , N - acetyl - L - cysteine treated group (NG) and zafirlukast treated group (ZG). Lipopolysaccharide (LPS,1 ml/kg) and saline (1 ml/kg) was injected into 1G and CG group, respectively. N - acetyl - L- eysteine (300 mg/kg) was given 2 rain before LPS (l ml/kg) injection in NG group while zafirlukast (20 mg/kg) was given 0.5 h and 1 h before LPS (1 ml/kg) injection, respectively. The pulmonary arterial pressure, PaO2, WBC, NO2^- concentration in the BALF, hung index (wet weight/body weight) and lung's wet/dry weight ratio (W/D) were detected in each group. Results:There were significant differences of the pulmonary arterial pressure and NO2^- concentration between NG and IG group (P 〈 0.05 ). PaO2 in two treated groups was significantly improved compared with IG group ( P 〈 0.05 ). Lung index and W/D ratio were significantly lower and WBC was higher in two treated groups than those in IG group (P 〈 0.05 ). Conclusion: N - acetyl - L - cysteine and zafirlukast can prevent rabbits from acute lung injury induced by the endotoxin.
