摘要
为了得到右旋的2-氨基丁酸,对2-氨基丁酸的拆分进行了研究.通过从猪肾中提取的氨基酰化酶选择性地水解(R,S)-2-N-乙酰氨基丁酸,得到了(S)-2-氨基丁酸,旋光度=+21.58℃(C=2,5mol/LHCl),收率为71.8%.试验结果表明最佳拆分条件为:底物浓度为0.1mol/L;底物与酶质量比为100:1;pH6.8~7.0;温度为37℃;反应时间为24h.
A study was made on the resolution of 2-aminobutyric acid in order to obtain dextrogyarte 2- aminobutyric acid. In our experiment, (S)-2-aminobutyric acid was resolved from (R, S)-2-aminobutyric acid by selective hydrolyzation of (R, S)-2-N-acyl aminobutyric acid which in the presence of acylase was derived from the kidney of pigs. The optical rotation= +21.58℃= 2,5N HCl) and the yield of (S)-2-aminobutyric acid is 71.8%. The experimental results showed that the best hydrolytic conditions are as follows: the concentration of the substrate is controlled to about 0. 1 mol/L;m(subsrate):m (enzyme) = 100 : 1 ; pH value of reaction system is 6.8 - 7.0 ; reaction temperature is 37℃ and reaction time is 24 h.
