在碱性条件下,刚果红与β-葡聚糖的结合具有高度专一性,能形成有色物质。基于此,结合流动注射技术建立了简便、快速测定麦芽汁及啤酒中β-葡聚糖的新方法。在优化的实验条件下,β-葡聚糖的线性范围为0.5~100.0mg/L(各线性段的相关系数r2≥0.9980,n=15),检出限(3σ)为0.2mg/L,回收率为95.0%~102.8%,相对标准偏差为1.23%(10mg/L的β-葡聚糖添加于麦芽汁中,n=16)及1.47%(10mg/L的β-葡聚糖添加于啤酒中,n=12)。该法具有较高的灵敏度、准确率和较强的抗干扰能力,每分钟可测17个样。将本法直接用于谷物制品(如麦芽汁和啤酒)中β-葡聚糖的分析,简便、快速,结果令人满意。
Under alkaline condition, the bond of Congo red and β-glucan had a high specificity and could form coloring matters. A new method for easy and rapid determination of β-glucan in grain products and beer combining with flow-injection technology was established based on this. Under optimized experimental conditions, the linear range of β-glucan was 0.5 - 100.0 mg/L ( correlation coefficient of each linearity range r^2≥ 0.998 0, n = 15 ), detection limit(3σ) was 0.2 mg/L, recovery rate was 95.0% - 102.8% and relative standard deviations(RSD) were 1.23% (10 mg/L β-glucan added into wort, n = 16)and 1.47% (10 mg/L β-glucan added into beer, 4n = 12 ) respectively. This method was feathered of high sensitivity and accuracy rate as well as strong anti-interference capability. It could measure 17 samples per minute and could be directly used for analysis of β-glucan in grain products, i.e. wort, beer or feed samples with easy, rapid and satisfied results.
