摘要
从实验室保藏的韦兰胶生产菌中筛选出作为育种的出发菌株,用紫外诱变得到系列突变体,经平板、摇瓶发酵及传代实验,筛选出韦兰胶生产菌株NC-2,并对其发酵培养基组分进行了正交优化实验,得到最优的发酵培养基配方为:蔗糖40g/L,酵母膏3g/L,K2HPO4.7H2O5g/L,MgSO4.7H2O4g/L,FeSO4.7H2O1mg/L,CaCl20.5g/L,产量达到15.20g/L,在出发菌株的基础上提高102.67%。
The Welan Gum producing strain was selected from the laboratory preservation strain as parent strain of breeding,many mutants were obtained by the Welan Gum producting strain with UV irradiation.The Welan Gum producing strain NC-2 was selected by plate, shake flask culture and generation experiments.Then the optimum fermentation medium was ensured by orthogonal design. It was showed that the prime fermentation medium contain.sucrose 40g/L, yeast extract 3g/L, K2HPO4 · 7H2O 5g/L, MgSO4·7H2O 4g/L, FeSO4·7H2O 1mg/L, CaCl2 0.5g/L,the yield of the NC-2 was 15.20g/L,more than that of the parent strain by 102.67%.
