摘要
目的探讨阿魏酸钠对大鼠单侧输尿管梗阻模型肾间质纤维化的影响。方法UUO大鼠给予阿魏酸钠100mg/(kg·d)腹腔注射,观察第14天血清肌酐、尿素氮的变化及肾间质纤维程度、ColⅠ、ColⅢ、t-PA、u-PA及PAI-1的表达。结果与UUO组相比,治疗组血清肌酐、尿素差异均无显著性,肾小管纤维化程度、PAI-1及Ⅰ、Ⅲ型胶原的表达显著降低,t-PA、u-PA的表达显著增加。结论阿魏酸钠可以通过下调PAI-1及上调t-PA、u-PA而减轻UUO术后大鼠肾间质纤维化。
[Objective] To investigate the effects of Sodium Ferulate on tubulointerstitial fibrosis of rat unilateral ureteral obstruction model. [Methods] Rats subjected to UUO were treatedwith daily peritoneal injection of 100 mg/kg of Sodium Ferulate. Creatinine, Urea nitrogen. The percentage of renal tubular lesion, interstitial fibrosis score were measured on 14th day after operation. The expression of t-PA, u-PA and PAI-1 were evaluated and compared. [Results] In comparison with UUO group, Creatinine, urea nitrogen were analogous, but the degree of renal tubulointerstitial fibrosis and interstitial expression of PAI-1, collagen Ⅰ and collagen Ⅲ were significantly reduced and interstitial expression of t-PA and u-PA were significantly increased. [Conclusion] Sodium Ferulate may play a protective role of the kidney by down-regulating PAI-1 and up- regulating t-PA and u-PA and alleviating renal interstitial fibrosis following UUO.
