摘要
以边沁桉Eucalyptus benthamii茎段的顶芽或腋芽为外植体开展了组织培养技术的研究,成功建立了边沁桉离体再生体系,为该物种进一步的快速繁殖提供了理论基础.结果表明:初接种芽诱导培养基以MS+BA0.2mg·L^-1最佳,诱导率为55.8%;最佳继代培养基为MS+BA0.5mg·L^-1+IBA1.0mg·L^-1+NAA0.2mg·L^-1,芽增殖系数为4.6,平均苗高为3.1cm;最佳生根培养基为1/4MS+IBA1.5mg·L^-1,生根率达93.3%,平均每株生根数为3.2条;试管苗练苗3~7d.移栽到25%糠壳灰+75%黄心土的基质,移栽成活率可达70%.
Using apical or axillary buds as explants, this paper conducted a study of Eucalyptus bentharnii tissue culture, establishing its in vitro regenerative system to quicken its proliferation. Results show that the optimum medium for initial-shoot induction is MS+ BA 0.2 mg · L^-1 with an induction rate of 55.8% that the optimum medium for subculture is MS+BA 0.5 mg· L^-1+IBA 1.0 mg· L^-1+NAA 0. 2 mg · L^-1 with a propagation coefficient of 4.6 and an average shoot height of 3.1 cm; and that the optimum medium for rooting is 1/4MS+IBA 1.5 mg · L ^-1 whose induction rate is up to 93.3% and whose averagenumber of roots is 3.2. Transplanted to pot soil mixed with bran ash and yellow subsoil (1 : 3) after hardening for 3-7 days, the cultured plantlets enjoyed a survival rate of 70%.
