摘要
[目的]探讨不同培养基配方诱导蝴蝶兰原球茎的效果及其诱导因素。[方法]以蝴蝶兰(f001和f006)试管苗根尖和叶为外植体,以MS和KC为基本培养基,分别添加不同浓度的6-BA、NAA和活性炭,各设置16个处理组进行原球茎诱导,研究其对诱导蝴蝶兰原球茎效果的影响。[结果]在KC+6-BA 1.0 mg/L+NAA 0.3 mg/L+AC 0.3%的培养基中,蝴蝶兰f006根段原球茎状体的诱导率可达20%;f001根段的诱导培养基配方采用KC+6-BA 1.0 mg/L+NAA 0.2 mg/L+AC 0.2%,其原球茎的诱导率达17%,f001叶片的诱导培养基配方采用KC+6-BA 4.0 mg/L+NAA 0.2 mg/L+AC 0.1%,其原球茎的诱导率达10%。[结论]该研究中所用的6-BA和NAA激素和活性炭浓度在MS和KC培养基上对蝴蝶兰原球茎的诱导影响均不显著;不同外植体在不同培养基中其适宜的外源激素浓度与组合也不同。
[Objective] The study aimed to discuss the effects of different media formulas for inducing Phalaenopsis protocorm and their inducing factors. [Method] With the root tip and leaf of Pbalaenopsis(fOOland fO06) test-tube shoots as explants,the MS and KC were taken as the basic medium adding different concn.of 6-BA and NAA and activated carbon resp.to set up the 16 treatments for protocorm induction and their effect on inducing Phalaenopsis protocorm were studied. [ Result] In the medium of KC + 6-BA 1.0 mg/L + NAA 0.3 mg/L + AC 0.3%, the protocorm induction rate of Pha/aenops/s f006 root segment was 20 %. When the induction mediums for f001 root segment was K C + 6-BA 1.0 mg/L + N AA 0.2 mg/L + A C 0.2 %, the protocorm induction rate was 17% .When the induction mediums for leaves was KC +6-BA 4.0 mg/L + NAA 0.2 mg/L + AC 0.1% ,the protoconn induction rate was 10%. [Conclusion] There was no significant effect of 6-BA and NAA hormones and the activated carbon concn, on the induction of Phalaenopsis proto- corm on MS and KC medium. The different explants on different medium had different exogenous hormone concn, and matching.
