摘要
利用三孢布拉氏霉发酵生产天然β-胡萝卜素。在30L发酵罐中,平均生物量31.3g干菌重/L发酵液和胡萝卜素1213.1mp/L;在3M^3发酵罐中,平均生物量38.0g干菌重/L发酵液和胡萝卜素1146.5mg/L。将中试样品经HPLC分析,在总色素中β-胡萝卜素占92~96%,其他类胡萝卜素占8~4%;在β-胡萝卜素中,反式异构体占90~95%,9-、13-、15-顺式异构体占10~5%。结晶β-胡萝卜素呈多种形状,但大多数为两端锥形的棱柱体。在胡萝卜素提取中,工艺和技术可靠,色素提取率90%以上。试验产品安全和卫生指标符合国家制定的标准。研究结果表明,利用该菌株可以实现工业化生产天然β-胡萝卜素。
Studies on production of natural β-carotene by fermentation with Blakeslea trisporaare reported in this paper. When five fermentation trials were conducted in a 30L fermentor, anaverage yield of carotenoids and biomass was 1213. ling and 31.1g dried mycilia / L fermentedbroth, respectively. An average of 1146.5mn carotenoids and 38.0g dried mycilia was obtainedin 3 fermentations performed in a 3M3 fermentor. Samples of mycilia and crystals preparedfrom 3M3 fermentor have been analyzed by RP-HPLC. The results demonstrated that β-carotene,as the predominant carotenoid, was 92-96%, other carotenoids were only 8- 4% of totalpigments. Of total β-carotene, the majority existed as all-trans isomer(90- 95%), but 15-cis,13-cis and 9-cis isomers (10- 5%) were detected in each sample. The crystallized β-caroteneappeares various shapes, but six-angle pyramid is the main shape. Downtreatment process forextracting pigments from mycilia has been designed, and > 90% of pigments has been regained.The preliminary product is coincident with the national standard of China both in safety andhealthy. These results demonstrate that B. trispora can be used in industry to produce naturalβ-carotene.
