摘要
目的探讨高脂膳食对脑老化及脑铁代谢的影响,为合理膳食预防脑老化提供科学依据。方法24只ICR小鼠分4组:脑老化模型组、高脂膳食组、脑老化+高脂膳食组和对照组。连续造模10w后,以RT-PCR法检测各组脑内铁调节蛋白-2(IRP-2)表达水平。结果脑老化组和高脂膳食组小鼠IRP-2表达水平均低于对照组且差异显著(P<0.05);高脂膳食组与脑老化模型组、脑老化+高脂膳食组间IRP-2表达无显著差异(P>0.05)。结论高脂膳食可降低小鼠脑内IRP-2表达,这可能是高脂膳食诱发脑老化及神经退行性变的机制之一。
Objective To investigate the effect of high fat diet as well as brain aging on iron metabolism in brain of mice and afford scientific evidence to rational diet and prevent brain aging. Methods ICR mice were randomly divided into four groups: brain aging model, high fat diet, brain aging + high fat diet and control groups. The brain aging mice model was established by subcutaneous injection of D-galactose (D-gal) at dose of 50 mg·kg^-1·d^1 for 10 w. The iron regulatory, protein-2 (IRP-2) mRNA level was determined by RT-PCR. Results The IRP-2 mRNA levels of mice in brain aging model and high fat diet groups were higher than those of control group (P〈0.05). But IRP-2 mRNA levels of mice in high fat diet group had no significant difference from those of brain aging and brain aging + high fat diet groups (P〉 0.05). Conclusions Similar to the effect of brain aging on IRP-2, high-fat diet can reduce the IRP-2 mRNA level in brain of mouse, which may be one of mechanisms for high fat diet inducing brain aging and neurodegeneration.
