摘要
通过水解低值海参——海地瓜(Acaudina molpadioides)的体壁蛋白,制备出具有显著抗高血压效果的水解产物。海地瓜的体壁蛋白先后被菠萝蛋白酶和alcalase蛋白酶水解,利用超滤技术,把水解产物分级成>2 kDa和<2 kDa两种组分。<2 kDa的组分具有很强的血管紧张素转换酶(ACE)抑制活性,其IC50值为0.615 mg/mL。肾血管性高压大鼠灌胃给药(<2 kDa的组分)1个月后,与模型组相比,其收缩压和舒张压均有明显下降,呈剂量依赖性,其中120μg/g剂量的降压效果与卡托普利10μg/g相当。
The purpose of the paper was to prepare hydrolysate of Acaudina Molpadioides body wall protein with high antihypertensive activity. Acaudina Molpadioides body wall protein was hydrolyzed sequentially with bromelain and alealase. The hydrolysate was fractionated into two ranges of molecular weight ( 〉 2 kDa; 〈 2 kDa) using an uhrafihration membrane bioreactor system. The fraction ( 〈 2 kDa) brought about a high ACE inhibitor), activity, with IC50 value of 0. 615 mg/mL. The fraction ( 〈2 kDa) was used as drinks administered to renal hypertensive rats (RHR) for 1 month. The systolic blood pressure and diastolic blood pressure of the RHR were significantly reduced compared with the model group in a dose-dependent manner, and the antihypertensive effect of the dosage ( 120 μg/g) is equivalent with eaptopril ( 10 μg/g).
