摘要
鱼鳞主要由表层的钙化层和内部的胶原纤维层构成,为提取鱼鳞胶原蛋白,首先要脱除鱼鳞表面的钙化层。本文主要研究鲤鱼鱼鳞在盐酸脱钙过程中的变化。鱼鳞在盐酸脱钙过程中外部形态首先发生变化,钙化层溶解变薄,鱼鳞的柔软性增加。随着脱钙过程的进行,盐酸逐渐渗透到鱼鳞内部。经过盐酸处理后鱼鳞的力学性质发生很大的改变,断裂强度大大降低,只有原始鱼鳞的1/3~1/4。说明鱼鳞中以羟基磷灰石为主要成分的无机物层对保护鱼鳞起到了很大的作用,也为鱼鳞坚韧的质构发挥了重要的作用。
Fish scales are composed of outer inorganic layer and inner organic fiber. In order to extract collagen from fish scales, inorganic layer should be detached first. The morphology changes of fish scales in the process of decalcification were studied. The calcific layer became thinner and thinner after fish scales were immersed in HCl solution. As the decalcification went on , the inner fibrillary plate structure became looser and more coarse. Elements such as Ca^2+ , Cl^- dissolving in HCI solution appeared in the energy spectrum of inner fish scale, which meant metal ions entered the inner fibrillary plate. Mechanical property of decalcified fish scale decreased to 1/3 - 1/4 of normal scale . It is concluded that the inorganic layer, mainly hydroxylapatite, plays a very important role in fish scale strength. The research work offered basic information for extraction of collagen from fish scales.
