原发性肝内胆管结石患者肝细胞中UDP-GT mRNA原位分子杂交

摘要 目的:观察原发性肝内胆管结石患者肝细胞内匍萄糖醛酸转移酶(UDP-GT)活性变化和该酶mRNA表达.方法:①用UDP-葡萄糖醛酸和2-氨基酚为底物,测定肝组织均浆UDP-GT活性.②地高辛标记cDNA探针与肝组织石蜡切片进行原位杂交,图像分析仪进行灰度测定,观察mRNA的表达.结果:患者组27%的患者UDP-GT活性降低,而正常对照组仅有14%的人降低;UDP-GT mRNA的表达虽有降低趋势,但统计学分析差异不显著.结论:原发性肝内胆管结石部分患者肝细胞内UDP-GT活性降低. The activities and mKNA expression of UDP-glucuronosyltransferase (UDP-GT) in hepatocytes were investigated in 18 patients with primary intrahepatic stone and 7 controls. Methods:①With UDP-glucuronic acid and 2-aminophenol as the substrate, UDP-GT activities were analyzed. The glucose 6-phosphatase, taken as a reference, was also measured.②By using in situ hybridization, the expression of UDP-GT mRNA was determined with digoxigenin-la-belecl UDP-GT cDNA probe. Results: Abnormal low activity of hepatic UDP-GT was found in 27% of 28 patients with primary intrahepatic stone, as compared with only 14% in 7controls. The UDP-GT mRNA expression was lower in the patients' hepatic tissues. But there was no significant statistic difference between the patient group and the controls. Con-elusion:Hepatoyte UDP-GT activity is discreased in patients with intrahepatic stone.
