摘要
目的:研究中华按蚊不同地理株基因组DNA的多态性.方法:应用RAPD技术,用20个随机引物(OPE01~OPE20)对江苏、福建、海南、贵州、陕西5个省区中华按蚊的基因组DNA进行了PCR扩增.结果:除OPE17,OPE18两引物对按蚊扩增片段较少外,其余18个引物均扩增出DNA片段.其中多数为2,3,4,5个地理株所共有的片段;少数为各个地理株独特的片段.5个地理株的PCR扩增条带在数量及片段长度上均有差别.结论:5个地区中华接蚊基因组DNA具有同源性,同时也存在着差异.
To study the DNA polymorphism of different strains of mosquito Anopheles sinensis. Methods: By using RAPD technology, genomic DNA of the mosquito Anopheles sinensis including strain Jiangsu, Fujian,Hainan,Guizhou, and Shaanxi was amplified with 20 random primers (OPE01-OPE20). Results: The primer OPE17 and OPE18 amplified few fragments,but the other primers amplified specific DNA fragments,most of them appeared in strain 2, 3, 4 and 5; and few of them was its specific fragments of each strains. The PCR amplified bands of 5 strains with different quantity and fragment length. Conclusion: Genomic DNA of 5 strains have homogenicity with differences among strains.
