摘要
目的:探讨外源性透明质酸(HA)抑制瘢痕形成的作用及其可能机制。方法:健康SD大鼠45只,复制背部创伤模型,随机分成2%HA治疗组(A组)、1%HA治疗组(B组)、PBS对照组(C组),观察创面大体形态,平均愈合时间和残留面积的大小;并测定愈合部位组织蛋白、羟脯氨酸(Hyp)、Ⅲ型前胶原(PCⅢ)的含量。结果:(1)3组的平均愈合时间为(15.2±2.0),(14.2±1.6)和(13.4±1.4)d,A、B两组与C组比较有显著性差异(P<0.05)。A、B两组的残留面积与C组比较有显著性差异,3组间比较也有显著性差异(P<0.05)。(2)Hyp和PCⅢ含量的测定:A、B两组Hyp含量的均值低于C组(P<0.01),而PCⅢ的含量却高于C组(P<0.01)。结论:(1)HA通过对细胞的调控作用,可有效地抑制胶原的合成,提高Ⅲ型胶原含量,使创伤愈合过程中瘢痕形成减少。
Objective: To evaluate the effects of hyaluronic acid(HA) on the inhibition of scar formation and their possible mechanisms. Methods: Forty-five healthy adult SD rats in a dorsal part trauma model were randomly divided into 2% HA treatedgroup (group A), 1% HA treatedgroup (group B), and PBS treatedgroup as a control group(group C). The process of healing was observed, the remained wound area was measured, and mean wound healing time was calculated. Furthermore, dynamic analysis of local tissue protein, Hyp and PC Ⅲ contents was performed. Results: (1) The remained wound area of control group was much smaller than that in treated group, and the mean value of wound healing time was(15.2±2.0), (14.2±1.6) and (13.4±1.4)d, respectively. (2) The contents of tissue Hyp were statistically lower in HAtreated groups, while those of PC Ⅲ were statistically higher as compared with. Conclusion: (1) The exogenous hyaluronan can effectively inhibit collagen synthesis and elevate tissue PCⅢ content. As a result, the scar formation is decreased. (2)HA efficacy presents a dosedependent manner.
