摘要
目的:研究谷氨酸对SW1990胰腺癌细胞系的增殖、凋亡及细胞毒性的影响及其可能的作用机制。方法:在体外用不同浓度的谷氨酸与人SW1990胰腺癌细胞作用后,用MTT比色法、BrdUrd渗入法和LDH法检测SW1990胰腺癌细胞的增殖能力、细胞分裂和死亡情况,并用激光共聚焦显微镜测定胞浆内游离钙离子浓度。结果:谷氨酸可以呈剂量依赖性地促进SW1990胰腺癌细胞的增殖(P<0.01),谷氨酸可以促进胰腺癌肿瘤细胞的分裂,减少其死亡(P<0.01),并且可以呈剂量依赖性地增加胞浆内游离钙离子浓度(P<0.01)。结论:谷氨酸可以促进肿瘤细胞的增殖,此作用可能是通过促进细胞的分裂、减少其死亡而实现的,且可能是通过增加胞浆内游离钙离子浓度介导的。
Objective To explore the effects of glutamate on the proliferation and apoptosis of pancreatic carcinoma cell line SW1990 and the possible mechanism of action of glutamate. Methods SW1990 cells were cultured in vitro and then treated with glutamate at different concentrations. The viability and apoptosis of the cells were detected by MTT assay, BrdUrd incorporation, and LDH release assay; the free intracellular concentrations of calcium ion were measured by confocal laser scanning microscopy. Results Glutamate stimulated the proliferation of pancreatic carcinoma cells in a concentration-dependent manner (P 〈 0.01 ). It increased cell division and decreased cell death (P 〈 0.01 ). Glutamate also enhanced the intracellular concentrations of calcium ion in a dose-dependent fashion (P 〈 0.01 ). Conclusion Glutamate can promote the proliferation of SW1990 cells via inducing cell division and decreasing cell death, which may be mediated by an elevation in the free intracellular concentrations of calcium ion.
