摘要
目的用高糖高脂肪饲料喂养大鼠,建立大鼠非酒精性脂肪肝模型。方法SD大鼠20只,适应3d,随机分为正常组和模型组,每组10只,♀♂各半。实验前2组大鼠的体质量、血清生化指标差别无统计学意义(P>0.05)。正常组的大鼠用基础饲料喂养,模型组的大鼠用高糖高脂肪饲料喂养。连续喂养30d,取血检测血清中生化指标;取肝脏检测肝组织中生化指标,并对肝脏作组织病理学观察。结果与正常组相比,模型组的大鼠摄食高糖高脂肪饲料30d,血清中TG、TC含量增加,ALT、AST活性升高(P<0.05);肝组织中TG、TC、MDA含量增加,SOD活力降低(P<0.05);肝脏组织病理学观察,正常组肝细胞无脂肪变,模型组肝细胞有明显的脂肪变。结论用高糖高脂肪饲料喂养大鼠,成功建立大鼠非酒精性脂肪肝模型。
Aim To construct a nonalcoholic fatty liver model by feeding rats with high glucose and high fat diet. Methods 20 rats are randomly divided into the normal group and model group after 3 days of adaptation. The difference in body weight and biochemical indicator in blood serum between the two groups was not statistically significant (P 〉 0.05 ). Then the rats in the normal group were fed with normal diet, and those of the model group fed with high glucose and high fat diet. After 30 days, the biochemical indicators in blood serum and in hepatic tissue were detected, and the pathological structure of the liver was observed. Results After being fed with high glucose and high fat diet for 30 days, compared with the normal group, the content of TG, TC and the activity of ALT, AST was in blood serum(P 〈0.05)in the model group; the contents of TG, TC, MDA were higher and the activity of SOD was lower in hepatic tissue (P 〈 0.05) ; The hepatocyte was without superfluous lipid droplet in the normal group, but was full of lipid droplet in the model group. Conclusion A nonalcoholic fatty liver model is successfully induced by feeding rats with high glucose and high fat diet.
