槐耳清膏诱导胃癌SGC-7901细胞凋亡的实验研究 被引量:13

摘要 目的研究槐耳清膏对胃癌SGC-7901细胞增殖抑制和凋亡诱导的作用,并探讨其作用机制。方法用终浓度为0、0.5、1、2、4、6 mg/mL的槐耳清膏和10μg/mL的5-氟尿嘧啶(5-Fu)作用于SGC-7901细胞,分别于24 h和48 h后采用MTT法检测细胞增殖抑制率;48 h后收集各组胃癌SGC-7901细胞,琼脂糖凝胶电泳检测DNA,流式细胞术检测细胞凋亡率,RT-PCR检测survivin mRNA表达。结果MTT法检测显示,槐耳清膏对胃癌SGC-7901细胞有增殖抑制作用,并存在浓度和时间依赖关系(P<0.05);槐耳清膏6 mg/mL组的抑制率48 h后达到(77.9±2.3)%,5-Fu组为(53.4±1.6)%,差异有统计学意义(P<0.05)。流式细胞术检测显示,槐耳清膏4 mg/mL组的细胞凋亡率最高,早期凋亡细胞达33.2%;6 mg/mL组早期凋亡细胞6.3%,而晚期凋亡细胞为19.9%。RT-PCR结果显示,槐耳清膏组胃癌SGC-7901细胞survivin mRNA表达下调。结论槐耳清膏在体外对胃癌SGC-7901细胞有明显的增殖抑制和凋亡诱导作用,初步推断槐耳清膏诱发胃癌细胞凋亡与其对survivin基因表达的抑制有关。 Objective To study the effects of Tramets robinophila on the proliferation inhibition and apoptosis induction of human gastric carcinoma SGC-7901 cells, and explore the mechanism. Methods Human gastric carcinoma SGC-7901 cells were treated with 0, 0.5, 1, 2, 4, 6 mg/mL Trainers robinophila and 10 μg/mL 5-fluorouracil (5-Fu), and the cell proliferation inhibition rates were determined by MTT assay 24 h and 48 h after treatment. Gastric carcinoma SGC-7901 cells were collected, DNA was isolated 48 h after treatment and was subjected to agarose gel electrophoresis. In addition, cell apoptosis rates were determined by flow cytometry, and RT-PCR was employed to detect the expression of survivin mRNA. Results It was demonstrated by MTT assay that the growth of SGC-7901 cells was inhibited by Tramets robinophila in a time-and dose-dependent manner(P 〈 0.05). The inhibition rate of the group of 6 mg/mL Tramets robinophila reached the peak 48 h after treatment, significantly different from that in the group of 10μg/mL 5-Fu [ (77.9 ±2.3)% vs(53.4 ±1.6)% ] (P 〈 0.05). It was revealed by flow cytometry that the cell apoptosis rate was the highest in the group of 4 mg/mL Tramets robinophila, and the early apoptosis rate was 33.2% , while the early apoptosis rate and late apoptosis rate were 6.3% and 19.9% , respectively for the group of 6 mg/mL Tramets robinophila. It was indicated by RT-PCR that the expression of survivin mRNA in SGC-7901 cells in the groups of Tramets robinophila was down-regulated. Conclusion Trametes robiniophila has inhibitory effects on SGC-7901 cell proliferation and can induce cell apoptosis, which may be related to the inhibition of the expression of survivin mRNA.
