全自动细胞处理系统制备冰冻红细胞的关键控制点及效果评价 被引量:7

摘要 目的应用ACP 215全自动细胞处理系统制备冰冻红细胞和冰冻解冻去甘油红细胞,观察冰冻解冻去甘油红细胞的质控指标,就其制备过程中的关键控制点、制备工艺及制备效果进行评价。方法随机取40袋2~6℃保存6d以内的悬浮红细胞用ACP 215全自动细胞处理系统制备冰冻红细胞,-80℃冰箱保存,37℃融化,再用ACP 215全自动细胞处理系统制备冰冻解冻去甘油红细胞。解冻红细胞洗涤前后留取样本检测冰冻解冻去甘油红细胞质量。结果红细胞回收率(86±5)%,游离血红蛋白浓度(0.27±0.16)g/L,体外溶血试验(6.76±6.9)%,甘油残余量(0.12±0.08)g/L,残留血小板量(0.03±0.02)%,残留白细胞量(0.24±0.25)%,细菌培养试验阴性。结论ACP 215全自动细胞处理系统可全程自动化无菌制备冰冻红细胞和冰冻解冻去甘油红细胞,方法操作简便,高效,产品质量稳定,更有效地保障了临床输血安全。 Objective To evaluate the key control point in preparing process, the preparing technology and preparing effect for preparing frozen red blood cells(FRBC) and frozen thawed deglycerolized red blood celIs(FTDRBC) with ACP 215. Methods At random way,40 Plastical bags suspended red blood cells(lu/bag) which stored within 6 days at 2-6 ℃ were prepared FRBC by the ACP 215 and were stored in refrigerator at -80℃. Then to thaw it at 37℃ and to prepar FTDRBC by the ACP 215. All samples of thawed deglycerolized red blood cells should be collected in order to check quality of FTDRBC. Results The recovery ratio of red blood cell was(86±5)%. The concentration of free hemoglobin was(0.27±0.16)g/L. The ratio of vitro hemolysis test was(6.76± 6. 9)%. The concentration of remnant glycerol was(0.12 ± 0.08)g/L. The ratio of remnant platelet was(0.03±0.02)%. The ratio of remnant leukocyte was(0. 24±0. 25)%. The bacterial culture test was all negative. Conclusion The ACP 215 can.prepare FRBC and FTDRBC full automatically and non-bacterial condition during whole process. The methods is easy, high efficiency and steady production quality. So as to ensure safety of clinical transfusion effectively.
