摘要
目的研究中药复方"石芍明目散"对糖尿病性白内障(DC)大鼠抗氧化损伤的影响。方法将Wistar大鼠100只随机分为对照组A、模型组B、石芍预防组C、石芍治疗组D、阳性对照组E,每组20只。A组常规饲养,其余4组喂高脂高糖饲料。B、C、D、E组大鼠采用链脲佐菌素(STZ)诱发糖尿病大鼠模型,A组腹腔注射等量生理盐水,B组以中药剂量等容的生理盐水灌胃,C组灌服石芍明目散治疗。3个月后处死(A、B组大鼠各处死一半)并检测A、B、C组大鼠晶状体中脂质过氧化终产物丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-px)的含量。同时开始给D、E组药物治疗。6个月实验结束处死并检测剩余各组大鼠。结果石芍明目散可明显降低大鼠晶状体中MDA含量,提高SOD、GSH-px活性,与模型组和阳性对照组比较,其差异有统计学意义(P<0.05)。结论石芍明目散能增强DM大鼠抗氧化能力,减轻晶状体的氧化损伤,延缓糖尿病性白内障病变的发展。
OBJECTIVE To study the anti-oxidative damage effects of Shishao Mingmu Powder, the traditional Chinese medicine compound formula on diabetic cataract (DC) rats. METHODS One hundred wistar rats were randomly divided into 5 groups: control group A,model group B, Shishao prevention group C, Shishao treatment group D, positive control group E. Each group has 20 rats. Group A was fed by conventional way. The remaining four groups were fed by high-fat and high-sugar feed. Rats in group B, C, D and E were streptozotocin (STZ)-induced diabetic rat model. Group A rats were injected isometric physiological saline. At the same time the stomach of the group C rats were filled with Shishao Mingmu Powder for prevention and treatment. Group B was filled physiological saline that had constant volume with the traditional Chinese medicine dosage. Half of the rats in group A and B were killed three months later. The content of the end-product of lipid peroxidation malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH - px) in the rats' crystal were tested. Group D and E were given drug treatment at the same time. Six months later at the end of the experiment the remaining rats were killed and tested. RESULTS Shishao Mingmu Powder could significantly reduce the dosage of the MDA and improve the activeness of the SOD, GSH-px in the crystal. The difference has statistical significance (P〈0.05-0.01) compared with model group and the positive control group. CONCLUSIONS Shishao Mingmu Powder could enhance DM rats' antioxidant capacity and reduce crystals' oxidative damage and delay the development of DC.
