摘要
目的探讨慢性间歇性缺氧以及复氧对大鼠学习记忆功能及海马CA3区胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)表达的影响。方法24只SD大鼠,分为空白组(UC组)、慢性间歇性缺氧组(CIH组)和复氧组(RH组),CIH组建立慢性间歇性缺氧模型,RH组前4周同CIH组,后4周正常饲养,所有大鼠在模型建立后进行Morris水迷宫测试后检测大鼠海马CA3区GFAP的表达。结果①Morris水迷宫逃避潜伏期成绩:CIH组>RH组>UC组(P<0.01,P<0.05);②穿越平台次数及跨越目标象限时间占整个游泳的时间百分率的比较:UC组>RH组>CIH组(P<0.01,P<0.05);③海马CA3区GFAP阳性产物COD值比较:CIH组>RH组>UC组(P<0.01)。结论①慢性间歇性缺氧可引起大鼠学习记忆功能的下降,复氧后大鼠学习记忆功能有所提高;②大鼠学习记忆功能的下降可能与海马CA3区GFAP的变化有关。
Objective To study the effects of chronic intermittent hypoxia and removal of hypoxia on learning and memory ability and glial fibrillary acidic protein (GFAP) expression in CA3 region of hippocampus in Sprague-Dawley rats. Methods A total of 24 Sprague-Dawley rats were randomly assigned to three experimental groups: unhandled control group (UC), chronic intermittent hypoxia group (CIH), and removal of hypoxia group (RH). Rats in CIH group and RH group underwent intermittent hypoxia for 8 h everyday. The duration of experiment was 4 weeks, and then those in RH group were fed normally in 4 more weeks. The learn- ing and memory ability of rats in each group was assessed with the Morris water maze test. GFAP expression in CA3 region of hippocampus was also investigated. Results (1) Morris water maze test learning scores: CIH group 〉 RH group 〉 UC group (P 〈0. 01, P 〈0.05); (2) Morris water maze test memory scores: The number of times of crossing the platform and the percentage of time spent on crossing the target quadrant to the total swimming time: UC group 〉 RH group 〉 CIH group (P 〈0.01, P 〈0.05) ; (3) GFAP expression in CA3 region of hippocampus : CIH group 〉 RH group 〉 UC group ( P 〈 0. 01 ). Ooflclusion Chronic intermittent hypoxia can attenuate the function of learning and memory in rats, but the function of learning and memory can be partly improved after removal of hypoxia, suggesting that the attenuated learning and memory in rats may be associated with the changes in GFAP expression in CA3 region of hippocampus in rats.
