摘要
以质粒pIJ702和pIJ699为载体,构建了两个带有透明颤菌(Vitreoscila)血红蛋白(VHb)基因(Vgb)的大肠杆菌-链霉菌穿梭质粒并转化E.coliHB101、变青链霉菌(S.lividans)TK64及蒽环类抗生素阿柔比星(阿克拉菌素)产生菌——加利利链霉菌(S.galilaeus)ATCC31133,经CO结合差光谱法、SDS-PAGE蛋白电泳分析表明Vgb在大肠杆菌和链霉菌中得到表达。通过改变培养条件研究溶氧对Vgb表达的影响,结果表明在低溶氧的情况下,VHb表达量增加。同时发现,在低氧浓度时,VHb的表达有利于链霉菌菌体的生长,其菌体生长量高于同样条件下的对照菌株(Vgb-)17%~29%。
Two shuttle plasmids containing Vitreoscilla hemoglobin (VHb) gene (Vgb) were constructed by cloning pUC19/Vgb in plasmid pIJ702 and pIJ699 and were introduced into E.coli HB101, Streptomyces lividans TK64 and Streptomyces galilaeus ATCC31133, respectively. The expression of VHb in the above mentioned hosts was verified by analysis of the CObinding difference spectra and the electrophoresis of SDSPAGE. It was demonstrated that under the condition of the lower dissolved oxygen concentrations the expression of VHb was increased and the growth of microbes was stimulated.
