摘要 目的探讨IKKβ和NF-κB p65在食管癌和癌前病变组织中的表达及意义。方法采用免疫组化PV-9000法检测正常食管上皮、基底细胞增生、间变和鳞状细胞癌中IKKβ和NF-κB p65的表达情况,以及在食管癌不同分化、淋巴结转移、浸润程度中的表达情况。结果IKKβ、NF-κB p65在不同组织中阳性表达率分别为:正常上皮(19.7%/12.8%)、基底细胞增生(46.7%/43.3%)、间变(76.0%/64.0%)和鳞状细胞癌(84.8%/75.8%),且随着食管病变进展,阳性表达率明显升高,正常上皮与鳞状细胞癌之间有明显差异(P<0.05);IKKβ、NF-κB p65的表达与食管癌组织的淋巴结转移和浸润程度呈正相关(P<0.05)。结论IKKβ、NF-κB p65在食管癌和癌前病变表达激活,可能与食管癌的发生发展及转移浸润密切相关。 Objective To evaluate the expressions and significance of IKKβ and NF-κB p65 in esophageal cancer and precancerous lesions. Methods Expressions of IKKβ and NF-κB p65 were determined by Immunohistochemistry (PV-9000) in normal esophageal epithelia, based cell hyperplasia, dysplasia and squamous cell carcinoma. Moreover, IKKβ and NF-κB p65 were also determined in esophageal squamous cell carcinoma of different degrees of differentiation, levels of metastasis and depth of invasion. Results Positive rates of IKKβ and NF-κB p65 in different tissues were as follows: 19.7%/12.8% ,46.7%/43.3% ,76.0%/64.0% and 84.8%/75.8% in normal esophageal epithelia (NOR) , based cell hyperplasia (BCH) , dysplasia (DYS) and squamous cell carcinoma (SCC) respectively. The pos- itive expression rates of IKKβ and NF-κB p65 increased with the lesion progressions. Moreover, there were significant differences between NOR and SCC (P 〈 0.05). And expressions of IKKβ and NF-κB p65 showed significant correlations with lymph node metastasis (P 〈 0.05) and depth of invasion (P 〈 0.05). Conclusion IKKβ and NF-κB p65 activate in esophageal cancer and precancerous lesions, which may correlate with pathogenesis, progression, metastasis and invasion of esophageal carcinoma.
