骨折合并脑外伤时骨愈合过程中神经生长因子作用的研究 被引量:9

The role of NGF during bone healing in fracture combined with traumatic brain injury

摘要 目的通过对脑外伤骨愈合过程中神经生长因子(NGF)合成变化的观察,探讨其在此过程中的作用,以此加深对骨生理学的认识,同时为干预骨愈合提供理论依据。方法72只大鼠随机分为正常组对照(N)组6只,脑外伤(TBI)组6只,骨折(F)组30只,骨折合并脑外伤(TBI+F)组30只。建立大鼠脑外伤模型,通过组织学观察及酶联免疫吸附法,研究骨折合并脑外伤时血清中NGF含量的变化。结果N组血清NGF OD值为0.041,F组手术后第3天血清NGF略有升高,1周时为最高,然后开始下降,4周时降至正常。TBI组和TBI+F组术后第3天血清NGF明显升高,1周后开始下降,5周时降至正常;其与TBI组或F组比较,差异有统计学意义(P<0.05)。结论NGF对促进骨质愈合可能有重要作用。 Objective To observe the changes of levels of nerve growth factor (NGF) during bone healing in fracture combined with traumatic brain injury, and to investigate its effect during the process in order to deepen the understanding for bone physiology, and to provide a theoretical basis for intervention of bone healing. Methods 72 rats were randomly divided into normal control group (group N, n = 6), traumatic brain injury group( TBI group, n = 6), fracture group( group F, n = 30), fracture combined with brain trauma group( TBI + F group, n = 30). The models of traumatic brain injury in SD rats were established, and histological observation and ELISA were used to observe the changes of NGF levels in serum during fracture combined with brain trauma. Results The serum OD value in NGF group was 0. 041, the levels of serum NGF in group F were slightly increased at 3 days after surgery, which would reach peak at one week after surgery, then which gradually decreased and reached normal levels on the 4th week. The NGF levels in TBI group and TBI + F group at 3 days after surgery increased obviously,which began to decrease at one week after surgery and reached normal levels on the 5th week,there were significant differences in the NGF levels between TBI + F group and TBI group or group F ( P 〈 0.05 ). Conclusion NGF may play an important role in promoting bone.
