摘要
目的进行栀子种质资源的遗传多样性研究。方法对12份栀子种质资源进行ISSR分析。利用POPGENE32软件分析遗传相似系数,UPGMA方法聚类,构建亲缘关系系统图。结果12个引物共扩增出126条谱带,其中多态性谱带92个,多态带百分率(PPB)73.02%;观测等位基因数(Na)1.730 2,有效等位基因数(Ne)1.464 3;Nei’s基因多样性(He)27.09%;Shannon’s信息指数(I)0.402 7。UPGMA聚类分析可将所有样品分为4类,聚类结果显示栀子种质亲缘关系与地理分布具有一定的相关性,并呈现出一定的地域性分布规律。结论栀子的遗传多样性水平较低,种质间的亲缘关系较近。
Objective To investigate the genetic diversity of Gardenia jasminoides Ellis. Method 12 specimens of germ plasma resources of Gardenia jasminoides Ellis were analyzed with inter-simple sequence repeats (ISSR) molecular markers.The systematic diagram of genetic relationships was established by POPGENE32 software and clustered by UPGMA method. Result Total 126 bands were amplified by 12 ISSR primers,in which 92 were polymorphic bands, the average percentage of polymorphie bands (PPB) was 73.02%, the observed number of alleles(Na) was 1.7302, effective number of alleles (Ne) 1.4643, Nei's gene diversity (He) 27.09%, and Shannon's information index(I) 0.4027. All the specimens might be grouped into four clusters by cluster analysis (UPGMA). Clustering results showed that germplasm relati jasminoides Ellis were related to the geographical distributions, revealing a onships of Gardenia tendency of regional distributions. Conclusion Different germplasm diversities of Gardenia jasminoides Ellis are lower and the relationships of Gardenia jasminoides Ellis are closer.
