摘要
目的研究金黄色葡萄球菌(简称金葡菌)α溶血素诱导脐静脉内皮细胞的凋亡效应。方法用不同浓度金葡菌α溶血素感染脐静脉内皮细胞,2、4、8和12h后,用流式细胞术检测细胞凋亡率。结果金葡菌α溶血素10、30、90和270ng/ml均可诱导脐静脉内皮细胞发生凋亡,以30ng/ml时诱导细胞凋亡能力最强,作用2h后细胞即发生凋亡,并随着诱导时间的延长,细胞凋亡率逐渐增加。结论金葡菌α溶血素可诱导脐静脉内皮细胞凋亡,并存在明显的时间、剂量效应关系,这可能是金葡菌L型垂直感染影响胎儿宫内发育的重要机理之一。
Objective To study the apoptosis of the human umbilical vein endothelial cells (HUVEC) induced by Staphylococcus aureus α-toxin. Methods The apoptosis of HUVEC infected by α-toxin was determined by the flow cytometry at different times after infection. Results S.aureus α-toxin at the dose of 10, 30, 90 or 270 ng/ml could induce apoptosis of HUVEC with a significant difference in the rates of apoptosis from those of the control, with maximal apoptosis rate at the 30ng/ml. Furthermore, the apoptosis of HUVEC was induced at 2 hours by S. aureus α-toxin at 30 ng/ml and the increase of apoptosis rates with the increase of infection time. Conclusion S. aureus α-toxin can induce the apoptosis of HUVEC with time-and dose-dependence, which may be the important mechanism by which S. aureus L- forms affect the growth and development of fetus in utero.
