High-Dose Estrogen Inhibits Prostate Development by Down-Regulating SHH Signaling in New-Born SD Rats

摘要 目的:探讨高剂量雌激素对新生SD大鼠前列腺形态发育的影响,及其与sonic hedgehog(SHH)信号通路之间的关系。方法:100只新生雄性仔鼠随机分为两组,出生后第0、1、3、5天分别给两组仔鼠皮下注射25μl玉米油和高剂量雌二醇(25μg/只,溶于25μl玉米油)。给药后于第1、6、10、20、30天取材前列腺,苏木精-伊红(HE)染色观察前列腺形态发育情况,免疫组织化学和RT-PCR分别检测SHH通路信号分子的表达。结果:高剂量雌激素组,新生SD大鼠前列腺腺体较小,腺管萎缩,发育明显受阻,在出生后第6、10、20、30天与对照组比较有显著差异;雌激素组SHHmRNA的表达在出生后第1、6、10、20、30天较对照组下调(P<0.05);雌激素组SHH信号通路的主要效应分子SHH、SMO、PTC和GLI-1的蛋白表达水平较对照组下调。结论:高剂量雌激素可能通过下调SHH信号通路,从而抑制新生SD大鼠前列腺的发育。 Objective : To investigate the effects of high-dose estrogen on the development of the prostate in new-born SD rats and its possible relationship with the sonic hedgehog (SHH) signaling pathway. Methods : One hundred new-born male SD rat pups were as- signed to an experimental group, subcutaneously injected with 25 μg Estradiol + 25 μl corn oil vehicle, and a control group, given corn oil alone, on postnatal day (PD) 0, 1, 3 and 5. The animals were sacrificed by decapitation and their prostates removed on PD 1, 6, 10, 20 and 30. The morphological changes of the prostate were observed by HE staining, and SHH signaling related molecules were detected by immunohistochemistry and reverse transcription PCR. Results: High-dose estrogen significantly inhibited the organogenesis of the new-born SD rat prostate and down-regulated the expressions of SHH signaling related molecules. Conclusion : Highdose estrogen may restrain the development of the prostate in new-born SD rats via inhibition of SHH signaling.
