摘要
目的:观察弓状核(ARC)毁坏后下丘脑-垂体-肾上腺-胸腺轴(HPAT)的改变。方法:大鼠出生后第2、4、6、8、10d皮下注射谷氨酸单钠(MSG)4mg/g,b.w。成年后检测HPAT轴的形态与功能。结果:MSG大鼠体重明显轻于生理盐水组,每100g体重所含垂体重量高于生理盐水组,肾上腺与胸腺重量未见明显差异;室旁核(PVN)促肾上腺皮质激素释放激素阳性细胞、正中隆起(ME)促肾上腺皮质激素释放激素阳性纤维及垂体前叶促肾上腺皮质激素分泌细胞数量明显多于生理盐水组,染色较深;血浆皮质酮及血浆促肾上腺皮质激素浓度高于生理盐水组。胸腺萎缩,淋巴细胞增殖反应、自然杀伤细胞细胞毒活性及白细胞介素2、γ干扰素的诱生能力明显低于生理盐水组。结论:1MSG-大鼠HPA轴功能亢进;2MSG-大鼠细胞免疫受抑。
AIM:To observe the changes of HPAT axis with lesion of arcuate nucleusMETHODS:Newborn Sprahue-Dawley rats were given subcutaneous injection of 4mg/g of monosodium glutamate (MSG) on days 2,4,6,8,10 of postnatal life, after they were adult, the morphological and functional changes of HPAT axis were examined. RESULTS:MSG-rats showed an increase of the weight of pituitary per 100gram body weight, while there were no difference in the weights of adrenal and thymus; the number of corticotropin releasing hormone (CRH) immunoreactive (IR) positive cells in paraventricular nucleus , CRH IR positive nerve teminals in ME and ACTH IR positive cells in pituitary increased significantly; baseline of plasma ACTH and corticosterone rose. Thymus atrophied with lymphocytes showing proliferative reactivity with natural killer cell cytotoxicity activity, productivity of IL-2 and interferon-γ declines significantly.CONCLUSION: MSG-rats showed hypofunction of HPA axis and the cellular immunofunction was inhibited.
