摘要
以小麦幼叶为材料,采用CTAB法1、CTAB法2、CTAB法3和NaOH法提取小麦基因组DNA,通过对小麦DNA提取方法的比较,为满足大批量材料进行SSR分子标记的检测寻找最优方法。结果表明,NaOH法提取DNA用时短、方法简单,但常有DNA样品扩增不出PCR产物的情况;CTAB法提取的DNA质量较高,而且CTAB法3省略了65℃温浴,并将两次氯仿∶异戊醇抽提改为1次抽提,既缩短了试验时间和降低了成本,提取的DNA又可以满足进行分子标记检测的需要。
In order to find out a better wheat DNA extracting method for the molecular marker analysis of large quantity samples, wheat leaves were used as experimental materials to study the DNA extracting methods such as CTAB l, CTAB 2, CTAB 3 and NaOH method. The resuhs showed that the NaOH method was very simple, but couldn't produce PCR. CTAB method reduced the cost and saved time for the experiment, improved the quality of DNA and simplified the extracting steps. The experiment indicated that CTAB 3 extracting method could save time and meet molecular marker analysis demand.
