摘要
目的探讨灯盏花素对大鼠急性脑缺血自由基水平和皮层脑源性神经生长因子(BDNF)表达的影响。方法采用线栓法制备大鼠大脑中动脉缺血-再灌注模型,分假手术组、生理盐水对照组和灯盏花素干预组,各组再分6、12、24、48h4个亚组。检测各组脑组织不同时间点过氧化物歧化酶(SOD)、丙二醛(MDA)含量,用RT-PCR和Western blot检测皮层BDNF mRNA和蛋白质表达变化。结果与对照组比较,干预组MDA含量降低,SOD活性增加。于48h时点,干预组BDNF mRNA表达较对照组明显增加。结论灯盏花素具有较明显的清除自由基作用,但对缺血后脑皮层BDNF蛋白的表达无显著影响。
Objective To study the effects of breviscapine on contents of free radicals and the expression of brain-derived neurotrophic factor (BDNF) in cerebral cortex after ischemia-reperfusion (I-R) injury of the middle cerebral artery in rats. Methods A total of 72 healthy male rats were randomly divided into sham operation group(S), control group(C) and breviscapine therapy group(B) with 24 rats each. Each group was further divided into 6, 12, 24, 48 h subgroups. The levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were examined. RT-PCR and Western blot were used to detect the expression of BDNF mRNA and protein, respectively. Results Compared with group C, the content of MDA was decreased and SOD activity was increased significantly in group B. The expression of BDNF mRNA at 48 h was significantly higher in group B than that in group C, but that of BDNF protein was not. Conclusion Breviscapine is a potent scavenger of free radicals but has no significant effect on the expression of BDNF protein after cerebral I-R injury in rats.
