摘要
红色果皮是杂草稻的基本特征之一,为了给杂草稻的分子鉴定提供依据,本研究根据已克隆的普通野生稻(Oryza rufipogen)红色果皮Rc与白色果皮rc等位基因第6外显子的差异,设计了InDel标记RID14。利用RID14标记对F2群体和江淮流域所收集到的24份红色果皮杂草稻、3份普通野生稻以及21份品种资源进行了标记基因型分析。研究表明,红色果皮均为非缺失带型,而白色果皮和紫色果皮为缺失带型,F2群体中杂合带型和非缺失带型均为红色果皮,而缺失带型为白色果皮。因此,RID14标记与白色果皮和红色果皮共分离,可以作为鉴定红色果皮杂草稻的分子标记。
Red pericarp is one of the basic characteristics of weedy rice (Oryza sativa L. f. spontaneous). In order to identify the weedy rice, an InDel marker, RID14, was designed based on the difference of extron 6 between red pericarp gene Rc and white pericarp gene rc. 24 weedy rice lines collected in 1950s and in recent years from the region of Yangtze and Huaihe River, 3 common wild rice (0. rufipogon) from Guangxi province, 21 germplasm lines and one F2 population derived from weedy rice were tested and genotype-analyzed with marker R/D14. The results indicated that RID 14 was cosegregated with the color ofpericarp in the F2 population and the germplasm, allowing to identify the homozygous red pericarp genotype, homozygous white pericarp genotype and heterozygous red pericarp genotype individuals. Therefore, the Indel marker RID14 is a perfect marker to identify the red weedy rice from commercial cultivated rice with white or purple pericarp.
