加味丹参饮对血瘀证家兔缺血再灌注损伤心肌细胞凋亡的影响 被引量:12

Influence of Jiawei Danshen decoction on cardiomyocyte apoptosis caused by myocardial ischemia reperfusion in rabbits with blood stasis syndrome

摘要 目的研究加味丹参饮对家兔缺血再灌注损伤血瘀证心肌细胞凋亡的影响,以探讨其对心肌缺血再灌注损伤(IRI)保护的作用机制。方法建立兔血瘀证IRI模型,以加味丹参饮对该模型进行干预,采用末段探针标记(TUNEL)法观察心肌细胞凋亡,采用放免法检测各组炎症因子(TNF-a和IL-2)水平变化。结果IRI组TNF-a、IL-2水平明显升高,心肌细胞凋亡显著增高,与空白组比较差异具有统计学意义(P<0.01);加味丹参饮组可以降低TNF-2、IL-2水平,抑制心肌细胞凋亡,与IRI组比较差异具有统计学意义(P<0.05或P<0.01)。结论加味丹参饮可以抑制IRI兔心肌细胞凋亡,降低炎症因子水平,可能是其保护心肌IRI作用机制之一。 Objective To study the influence of Jiawei Danshen decoction (JDSD) on eardiomyoeyte apoptosis and inflammatory factors of myocardial isehemia reperfusion (IRI) rabbits with blood stasis syndrome and investigate protective mechanism of JDSD on myocardial IRI. Methods The IRI model of blood stasis syndrome was intervened with the decoction, levels of eardiomyocyte apoptosis was evaluated with the method of terminal deoxynucleotidy transferase mediated nick end labelling (TUNEL), and the contents of tumor necrosis factor-alpha (TNF-α)and intedeukin-2 (IL-2) was detected by radio immune assay. Results Compared with the IRI group, the contents of TNF-α and IL-2 were decreased significantly and cardiomyocyte apoptosis was decreased significantly in JDSY group (P〈0.01 or P〈0.05). Conclusion JDSY can inhibit eardiomyocyte apoptosis and decrease inflammatory factors in rabbits with myocardial IRI, which may be the mechanism to prevent from myocardial IRI.
