摘要
目的:测定乌头注射液的药动学参数,为临床合理用药提供科学依据。方法:以小鼠的急性死亡率为指标,先后两次ip给药,测定首次给药后不同时间药物的体存量,再进行房室模型拟合和药动学参数估算。结果:小鼠ip给药后,药物的消除呈一级动力学过程,并符合一室模型。其表观消除速率常数k为0.00117min-1,其表观半衰期为59.23min。结论:目前临床一日2次给药间隔时间太长,如果每8h给药1次,首次倍量。
OBJECTIVE: To determine pharmacokinetic parameters of Wutou injection in mice, and provide scientific basis for its rational use. METHOD: According to the index of acute mortality of mice, Wutou injection was administered ip twice. We determined the remanent dose in mice in different times after the first ip, then the apparent parameters of pharmacokinetics were estimated. RESULTS: The first order kinetics and one compartment model were noted on elimination of the drug, after Wutou injection ip in mice. The apparent elimination rate constant k was 0.001 17 min -1 . The apparent half life time t 1/2 was 59.23 min. CONCLUSIONS: At present Wutou injection is administered twice a day, the intermission time is too long. If it is given every 8 h and double dose is prescribed in the first time, the curative effect may be better and the toxicity lowered.
