摘要
目的观察葶苈生脉饮对压力负荷增加致充血性心力衰竭(CHF)大鼠心肌组织基质金属蛋白酶-9(MMP-9)及基质金属蛋白酶组织抑制因子-1(TIMP-1)表达的影响。方法建立腹主动脉缩窄所致充血性心力衰竭大鼠模型,随机分为假手术组、模型组、葶苈生脉饮高剂量组、葶苈生脉饮低剂量组、阳性药(心宝丸)对照组。使用MS4000 U生物信号定量记录分析系统检测各组大鼠的LVSP、LVEDP、±dp/dtmax变化;采用免疫组化法观察各组大鼠心肌组织MMP-9、TIMP-1的表达情况。结果模型组大鼠LVEDP、-dp/dtmax升高,MMP-9表达增强(P<0.05或P<0.01);LVSP、+dp/dtmax降低,TIMP-1表达减弱(P<0.05或P<0.01)。各给药组与模型组比较,LVEDP、-dp/dtmax降低,MMP-9表达减弱(P<0.05或P<0.01);LVSP、+dp/dtmax升高,TIMP-1表达增强(P<0.05或P<0.01);其中高剂量组明显优于低剂量组和心宝组(P<0.05或P<0.01)。结论葶苈生脉通过调控MMP-9和TIMP-1的表达,可能是其逆转心室重塑、增强心脏功能、治疗CHF的作用机制之一。
Objective To observe the effects of Tinglishengmaiyin on the expressions of matrix metalloproteinases-9(MMP-9) and tissue inhibitor of metalloproteinases(TIMP-1) in the myocardiac tissue of rats with congestive heart failure(CHF) induced by over loaded pressure.Methods The model rats with CHF were established by suprarenal aortic constriction operation and divided into 5 groups randomly: sham operated,model control,Tinglishengmaiyin high,low dose groups and Xinbaowan group.The MS4000U biological signal collecting system was used to record and analyze the changes of LVSP,LVEDP and±dp/dtmax.Immunohistochemical method was used to observe the expressions of MMP-9 and TIMP-1.Results In model control group,the LVEDP,-dp/dtmax,the expression of MMP-9 increased(P0.05 or P0.01) and the LVSP,+dp/dtmax,the expression of TIMP-1 decreased(P0.05 or P0.01).Compared with model control group,LVEDP,-dp/dtmax,the expression of MMP-9 decreased(P0.05 or P0.01),LVSP,+dp/dtmax,the expression of TIMP-1 increased(P0.05 or P0.01) in the treatment groups.High dose group was significantly superior to low dose group and Xinbaowan group(P0.05).Conclusions Tinglishengmaiyin can adjust the expressions of MMP-9 and TIMP-1,which may be one of the mechanisms of reversing myocardial model and improving heart function to treat CHF.
