摘要
利用原代培养的小鼠肝细胞,研究烟酰胺(Nieotinamide,NA)对细胞能量代谢的影响。以不同浓度的NA(0—500μmol/L)干预24h后,检测细胞和培养介质中一氧化氮(NO)含量,一氧化氮合酶(NOS)活力及三种能量代谢指标乳酸(LD)、乳酸脱氢酶(LDH)和三磷酸腺苷酶(ATPase)。结果表明,随着NA浓度的增加,其对肝细胞能量代谢的促进作用逐渐上升,当浓度为200μmol/L时达到高峰,即200μmol/LNA的促进效果最为显著,而当NA浓度达到500舢oL/L时,肝细胞能量代谢水平反而低于对照组。表明一定浓度范围内烟酰胺具有促进肝细胞能量代谢的作用。
To investigate the effect of nicotinamide (NA) on energy metabolism of mice hepatocytes in vitro, hepato- cytes were cultured in the presence of 0,100,200,300,400 and 500 μmol/L of NA for 24 h,then nitric oxide (NO),nitric oxide synthase (NOS) and three indexes lactic acid(LD), lactate dehydrogenase(LDH) and adenosine triphosphatase (AT- Pase ) were assessed by kits. Results showed that 200μmol/L instead of 500μmol/L NA significantly stimulated energy metabolism of mice hepatocytes, which suggested that NA can promote energy metabolism of mice hepatocytes in vitro.
