粉叶爬山虎红色素的提取纯化工艺研究 被引量:2

摘要 本文采用酸性乙醇溶液提取粉叶爬山虎果皮中红色素,比较了九种大孔吸附树脂对红色素静态、动态吸附和解吸实验,筛选出红色素纯化的最佳树脂,并用薄层色谱法对红色素进行了单体分离。结果表明:LSA-8大孔吸附树脂纯化红色素效果较好,确定了吸附和洗脱的最佳工艺参数为:上柱液pH6.5~7.5,样液质量浓度0.56mg/mL,吸附流速2.0mL/min;以60%乙醇为解吸剂,洗脱液流速1.0mL/min;通过薄板层析(TLC)研究,得其TLC最佳展层剂是:V(水):V(甲酸):V(乙酸乙酯)=3:2:10,分离红色素,得到4种单体成分。 In this paper, red pigment was extracted from Parthenocissus thomsonii plach using acidic ethanol solution. By comparison of static and dynamic adsorption and desorption of red pigment on macroporous adsorbent resins, the optimum adsorbent resin was determined as LSA-8 macroporous adsorbent resins. The optimum conditions for adsorption and elution of the red pigment were: pH value of sample solution being of 6.5-7.5, sample concentration being of 0.56 mg/mL, the flow rate of adsorption being of 2.0 mL/min, the desorption solvent being of 60% ethanol, and the flow rate of eluent being of 1.0 mL/min. Four monomers were isolated from the red pigments by thin layer chromatography(TLC), and the optimum ratio of the developing solvents was H2O: formic acid : Ethyl acetate of 3:2:10.
