黑芝麻提取物促B16黑素瘤细胞黑素合成及其机制的研究 被引量:8

The Effect of Water Extract from Semen Sesami Nigrum on Melanogenesis of B16 Melanoma Cells

摘要 目的研究黑芝麻的水提取物对B16黑素瘤细胞系酪氨酸酶活性、黑素合成及相关基因和蛋白表达的影响,探讨黑芝麻促进黑色素合成的可能机制。方法选择3种浓度(0.5,1,2 mg/ml)的黑芝麻水提物作用于B16黑素瘤细胞72h,观察其对黑素细胞的增殖、酪氨酸酶活性和黑素含量的影响。免疫印迹法和半定量逆转录聚合酶链式反应(RT-PCR)分别检测药物作用48 h前后酪氨酸酶(Tyrosinase)、小眼相关转录因子(microphthalmia associated transcription factor,MITF),酪氨酸酶相关蛋白酶1(TRPl)和酪氨酸酶相关蛋白酶2(TRP2)蛋白和mRNA表达水平的变化。结果3种浓度的黑芝麻水提物可轻微促进B16细胞的增殖(P>0.05);试验浓度范围内可明显促进B16细胞黑素合成和酪氨酸酶活性增加(P<0.05),并以浓度依赖方式促进酪氨酸酶和MITF的蛋白和mRNA表达(P<0.05),但对TRP-1、TRP-2的表达几乎没有影响(P>0.05)。结论黑芝麻水提物在体外能直接刺激B16黑素瘤细胞黑色素的合成,上述变化可能通过促进酪氨酸酶和MITF的基因转录和蛋白表达作用来完成。 Objective To study the effect of water extract from Semen Sesami Nigrum (WES) on cell viability, tyrosinase activity and melanogenesis of B16 murine melanoma cells and to clarify the mechanism of its stimulating effect on melanogenesis. Methods Three different concentrations of WES were incubated with B16 cells for 72h and then cell viability, melanin content, tyrosinase activity were respectively measured. The protein and mRNA expression of melanogenesis key enzymes were investigated by western blot and RT - PCR after 48 h treatment of WES. Results Three different concentrations of WES slightly increased cell viability (P 〉 0.05). In the range of experimental dose, WES increased the activity of the tyrosinase and melanin content significantly in a dose -dependent manner( P 〈 0.05). The increasing mRNA and protein expression of TYR and MITF were also observed in a dose - dependent manner( P 〈 0.05 ). Almost no effect was observed on the expression of TRP1 and TRP2 (P 〉 0.05). Conclusion WES can directly stimulate melanogenesis in B16 melanoma cells, which may be induced by increasing expression of TYR and MITF on mRNA and protein levels.
