摘要
目的探讨解毒化瘀颗粒抑制急性肝衰竭小鼠肝细胞凋亡的作用及可能的机制。方法将昆明小鼠随机分为空白组、模型组、解毒化淤颗粒组、安宫牛黄丸组、乳果糖组,用D-GalN+LPS腹腔注射构建急性肝衰竭的小鼠模型,观察各组小鼠存活率,SYBR荧光实时定量PCR法检测肝细胞内天冬氨酸特异的半胱氨酸蛋白酶-3(Caspase-3 mRNA)的表达。结果成模后48 h存活率解毒化淤颗粒组高于其他药物组;促凋亡因子Caspase-3在解毒化淤颗粒组肝组织中表达量低,而在其他药物干预组及模型组中则高表达。结论急性肝衰竭中Caspase-3表达与肝细胞凋亡程度有相关性,解毒化淤颗粒能下调内毒素肝损伤肝细胞Caspase-3表达并抑制其凋亡效应,降低肝衰竭小鼠死亡率,提示解毒化淤颗粒抑制肝细胞凋亡有可能是其防治急性肝衰竭的机制之一。
Objective To investigate the effect of Jieduhuayu granule on hepatocyte apoptosis in acute liver failure mice and its possible mechanism. Methods Kunming mice were randomly divided into blank control group, model group, and Jieduhuayu treating group. Acute liver failure was built with D - GalN + LPS intraperitoneal injection to the mice. The survival rate was observed and the expression of Caspase - 3mRNA was tested with SYBR fluorescence real - time quantitative PCR assay. Results After 48 h, the survival rate in Jieduhuayu treating group was higher than that of other drug treating group ; at the same time pro - apoptotic factor,Caspase- 3 expression in liver tissue is low in the Jieduhuayu particle group but high expression in other drug treatment groups. Conclusion The Caspase - 3 expression is related with hepatocyte apoptosis in acute liver failure. Jieduhuayu treatment granule can reduce liver cell injury by through inhibiting the expression of Caspase - 3, and reducing mortality rate of liver failure mice.
