摘要
本文采用制备大鼠肺条件培养基进行单层软琼脂法培养大鼠骨髓造血祖细胞(GM—CFU),进行定量研究。结果显示肺条件培养基的刺激因子(GM—CSF)活性好,15%浓度的CM刺激2.5月龄大鼠骨髓每4×10~5个有核细胞中形成细胞集落92.5±23.8。定量研究结果表明,老年大鼠的GM—CFU明显下降,如青年大鼠(2.5月龄)的GM—CFU为92.5±23.8,而老年大鼠(24月龄)的GM—CFU则为25.4±9.6。进一步观察发现有随年龄增长而下降的趋势,提示老化的机体其骨髓造血功能是低下的。
The paper present a puantitative method for the hemopoietic function in Rat. The conditioned medium (CM) of rat lung was prepared, and Soft-agar culture in vitro of hemopoietic progenitor (GM-CFU) of bone marrow was used. The activity of colony stimulating factor(CSF) in CM was high, and 92.5±23.8 colonies formed from 4×10~5 nucleated cells of bone marrow as 15% CM was addad.We found that the GM-CFU in aging rats decreased. The GM-CFU in young rats (2.5 months) was 92.5±23.8, but that in aging rats (2 years old)was 25.4±9.6 and furthermore, it was observed that the GM-CFU reduced with age. The GM-CFU in one year old was 37.0% of that in young rats, in 1.5 years old rats was 35.7%, in two years old rats was 27.2%. Obviously, The hemopoietie function in aging organism wasd decreased.
