摘要
目的:优化金银花药材中木犀草苷的含量测定方法。方法:采用高效液相色谱法,以苯基硅烷键合硅胶为填充剂,色谱柱为Agilent ZORBAX SB-phenyl(4.6mm×250mm,5μm)柱,流动相为乙腈-0.5%醋酸溶液系统,梯度洗脱,流速1mL·min-1,检测波长350nm,柱温25℃,进样量10μL。结果:木犀草苷进样量在0.09~0.91μg范围内与峰面积呈良好的线性关系(r=0.9999);平均回收率(n=6)为103.1%,RSD为2.1%;检测限(S/N=3)为3.48ng,定量限(S/N=10)为8.70ng。结论:本法简便准确,可用于金银花药材中木犀草苷的含量测定。
Objective :To develop an HPLC method for the determination of luteolin - 7 - O - glueoside in Flos Lonicerae Japonicae. Method:An Agilent ZORBAX SB -phenyl column(4. 6 mmx 250 mm,5 μg m) was adopted; The mobile phase was acetonitrile -0.5% acetic acid with gradient elution mode at a flow rate of 1 mL · min-1 ; The detecting wavelength was 350 nm, the column was 25 ℃, and the injection volume was 10 μL. Results :The cal- ibration curve was linear in the range of O. 09 -0. 91 p,g of lutcolin -7 -O -glucoside with correlation coefficient of O. 9999;The average reeovery(n =6) was 103.1% with RSD of 2. 1% ;The limit of detection(S/N =3) was 3.48 ng and the limit of quantification( S/N = 10) was 8.70 ng. Conclusion: This method is accurate, and can be used to determine the content of luteolin - 7 - O - glucoside in Flos Lonicerae Japonicae.
