直肠癌中肿瘤相关巨噬细胞、基质金属蛋白酶-9与微淋巴管生成关系

The Relationship among Tumor-associated Macrophages、 Matrix Metalloproteinase-9 and Lymphangiogenes in Rectal Carcinoma

摘要 目的:研究直肠癌的微淋巴管生成情况、肿瘤相关巨噬细胞(TAM)计数与基质金属蛋白酶(MMP)-9表达的关系,探讨TAM与MMP-9在直肠癌微淋巴管生成中的作用。方法:通过免疫组化法测定41例直肠癌标本微淋巴管密度(LMVD)、TAM密度及MMP-9的表达,并分析LMVD、MMP-9与直肠癌其它临床病理学因素间的关系。结果:①直肠癌组织LMVD、TAM密度、MMP-9的表达明显高于正常组织(P<0.05)。②TAM浸润与MMP-9表达和肿瘤微淋巴管生成相关(P<0.05)。③三者与肿瘤分化程度、Dukes分期、淋巴浸润和淋巴结转移等有不同程度的相关性。结论:TAM与MMP-9、直肠癌微淋巴管生成在直肠癌转移中起着重要的作用,MMP-9很可能是肿瘤微淋巴管生成的始动因子之一。 Objective: To investigate the relationship among lymphangiogenes, tumor-associated maerophages(TAM) and matrix metalloproteinase-9(MMP-9)in rectal carcinoma, and to discuss the pivotal roles of TAM and MMP-9 in lymphangiogenes of rectal carcinoma. Methods: Forty-one resected tumor specimens were obtained from patients and the immunohistochemical staining was used for detecting the expression of MMP-9, lymphatic microvessel density(LMVD) and TAM density, and the statistics was used for analyzing the inter-relationships and the correlation among them and clinicopathological parameters. Results: LMVD, TAM density and the expression of MMP-9 in rectal carcinoma were significantly higher than those in normal tissues( P 〈 0.05); Correlations were found among TAM invasion, the expression of MMP-9 and the lymphangiogenes( P 〈 0.05); LMVD, TAM density and the expression of MMP-9 correlated with histologic type, infiltration extent(Dukes), lymphatic invasion, lymph node metastasis at different degrees. Conclusion: TAM density, the expression of MMP-9 and lymphangiogenes may be critical in rectal carcinoma metastasis. MMP-9 supposes to be one of the initiating factors of carcinoma lymphangiogenes.
