循环高速逆流色谱从曼地亚红豆杉枝叶提取物中分离紫杉醇和三尖杉宁碱 被引量:6

Separation of paclitaxel and cephalomanine from extracts in Taxus media branches and leaves by recycling high-speed counter-current chromatography

摘要 目的研究曼地亚红豆杉枝叶提取物中分离紫杉醇及其类似物三尖杉宁碱的方法,提高紫杉醇的回收率及质量分数。方法曼地亚红豆杉枝叶提取物先经Al2O3柱纯化后,再以正己烷-醋酸乙酯-甲醇-乙醇-水(5∶7∶5∶1∶6.5)为溶剂系统,利用循环高速逆流色谱分离紫杉醇和三尖杉宁碱。结果经循环高速逆流色谱循环一次后,三尖杉宁碱与紫杉醇即达到基线分离,紫杉醇质量分数达98.2%以上,回收率83.0%。结论循环HSCCC法为分离紫杉醇和三尖杉宁碱提供了一条简便、快速的方法,使制备高质量分数、高回收率的紫杉醇成为可能。 Objective To study the method for separating paclitaxel from the extracts in Taxus media branches and leaves and cephalomanine, and to increase recovery rate and content of paelitaxel. Methods The extract in T. media branches and leaves was purified by Al2O3 firstly. Then the recycling high-speed counter-current chromatography (HSCCC) was used to separate paclitaxel and cephalomanine from the crude sample with a solvent system composed of n-hexane-ethyl acetate-methanol-alcohol-water (5 : 7 : 5 : 1 : 6.5). Results Paclitaxel and cephalomanine were completely separated by recycling HSCCC prepa- ration with one recycle. The content of paclitaxel was over 98.2% and the recovery rate was 80.3%. Conclusion The recycling HSCCC provides a convenient and rapid way to separate paclitaxel and cephalomanine which could make it possible to prepare paclitaxel with high content and recovery rate.
