Comparative Study on Inflammatory Cytokines in Bleomycin Induced Pulmonary Fibrosis in Rats

摘要 目的:了解肺纤维化大鼠肺局部细胞因子水平及其与肺泡巨噬细胞的关系。方法:对比观察支气管肺泡灌洗液(BALF)和肺泡巨噬细胞(AM)培养上清肿瘤坏死因子-α(TNF-α)、血小板源生长因子(PDGF)的动态变化。二种细胞因子检测分别采用ELISA和生物活性法。结果:①BALF中TNF-α于第1d增高,第3d达高峰,其高峰时间先于AM源TNF-α;BALF中TNF-α与其中中性粒细胞数呈正相关;②BALF中PDGF与AM源PDGF同步于第7d达高峰,与AM总数呈正相关。结论:AM释放高水平TNF-α、PDGF;BALF中也存在TNF-α、PDGF,且呈动态变化;BALF中TNF-α可能还来自AM外的其它细胞,如PMN,而BALF中PDGF则可能主要来源于AM。 Objective:The purpose of this article was to delineate the changes of cytokines in lung foci and the correlationship among alveolar macrophage (AM) derived TNF α, platelet derived growth factor (PDGF), and the same cytokines in pulmonary focuses. Methods:We compared the AM derived TNF α, PDGF and BALF TNF α and PDGF; TNF α and PDGF in AM supernatant and BALF were measured with ELISA and bioassay respectively. Results: (1) TNF α in BALF significantly increased on 3rd ady and decreased instantly to normal levels after 7th day; AM derived TNF α increased on 3rd day, peaked on 7th day and then remained at higher level till the 28th day. (2) The peak of BALF TNF α preceded that of AM derived TNF α. (3) A good positive correlation was found between BALF TNF α and neutrophil percentage. (4) BALF PDGF peaked on 7th day and gradually decreased to normal, which was parallel to the changes of AM derived PDGF; PDGF in BALF correlated well with the total AM count. Conclusions: TNF α PDGF in AM supernatant and BALF exhibited consecutive changes in this model, other immune effector cells might also contribute to the presence of TNF α in BALF besides AM. PDGF in BALF might mainly originated from AM.
