热应激对大鼠睾丸iNOS表达的影响及其意义 被引量:2

The significance and effect of heat stress on the expression of iNOS in the rat testis

摘要 目的研究热应激对大鼠睾丸诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)表达的影响及其意义。方法42d龄SD雄性健康大鼠12只建立单侧隐睾作为热应激处理的模型,术后第7d处死大鼠留取双侧睾丸,采用RT-PCR及免疫组化SABC法分别检测生精细胞iNOSmRNA和蛋白表达的变化,TUNEL法检测生精细胞凋亡。结果热应激处理后生精上皮内可见iNOS强阳性表达,而对侧睾丸生精上皮内无iNOS表达;热应激处理后睾丸iNOSmRNA也较对侧睾丸增强,生精细胞凋亡指数明显高于对侧睾丸,差异均有显著性意义(P<0.05)。结论热应激导致生精上皮内iNOS表达增强,生精细胞凋亡增加,iNOS参与介导生精细胞凋亡。 Objective To investigate the significance and effect of heat stress on the expression of iNOS in the rat testis Methods Unilateral cryptorchidism was surgically induced in the 42-day old rats as the model of heat stress. The expression of iNOS was detected by RT-PCR and immunohistochemieal SABC assays on the 7th day after the operation. The apeptosis of spermatogenie cell was detected by terminal-deoxynueleofidyl transferase mediated d-UTP nick end labeling (TUNEL). Results In spermatogenic epithelium of the contralateral testis there was no iNOS expression which could be detected in the cryptorchidism by immunohistochemieal assay. Compared to the contralateral testis the apeptosis index(Al) of spermatogenic cell and the expression of iNOS mRNA in the cryptorehid testis increased significantly(P〈0.05). Conclusions Heat stress can enhance the expression of iNOS in spermatogenic epithelium and the apeptosis of spermatogenie eeU which is partly mediated by iNOS.
