酶解辅助水法提取土茯苓总黄酮工艺条件研究 被引量:1

Study on the Extraction Process of Total Flavonoids from Smilax Glabra by Enzyme-assisted Hydrolysis

摘要 采用了酶解辅助水法提取土茯苓总黄酮.以芦丁为标准品,用铝离子显色、紫外-可见分光光度法测定总黄酮含量.以总黄酮提取率为考察指标,通过单因素实验探讨酶用量、固液比、提取温度、提取次数对总黄酮提取率的影响,优化提取条件.结果表明最佳提取工艺条件为:酶用量2.0%、固液比例1:8、提取温度100℃、提取3次.最佳条件下,粗提物干粉得率为14.0%,干粉总黄酮含量为4.1%,总黄酮提取率为0.57%. The total flavonoids was Extracted from Smilax glabra by a enzyme-assisted hydrothermal method; Using Rutin as standard, the content of the total flavonoids was measured by ultraviolet and visible spectrophotometry with aluminum ion colour. The extraction ratio of total flavonoids was investigated as a main target. The effects of amount of enzyme, the solid-liquid ratio, extraction temperature and extraction times on the total extraction ratio of flavonoids were discussed by means of the single-factor experiment to optimize the extraction conditions. The results showed that: for the best extraction conditions the enzyme dosage is 2.0% with the solid-liquid ratio being 1:8 and extraction frequency being 3 times at 100 ℃. In the best conditions, the powder ratio of crude extraction is 14.0%, and the content of total flavonoids in this powder is up to 4.1% with the extract ratio of total flavonoids being 0.57%.
