摘要
目的从鲍鱼内脏中提取活性鲍鱼内脏蛋白多糖(AVPF-I),检测AVPF-I的抑瘤作用及对荷瘤小鼠免疫功能的影响。方法采用H22移植瘤模型,设生理盐水空白对照、环磷酰胺阳性对照和AVPF-I组,测定各组小鼠瘤重、脾脏指数,MTT法测定淋巴细胞增殖及NK细胞杀伤活性,中性红吞噬法测定腹腔巨噬细胞吞噬活性,ELISA试剂盒检测TNF-α,IL-1和IFN-γ含量。结果AVPF-I能够显著抑制肿瘤生长(P<0.01),抑瘤率达62.37%;与阴性对照组比较,实验组荷瘤小鼠的脾脏指数显著提高(P<0.01),T淋巴细胞增殖作用、NK细胞的活性、巨噬细胞的吞噬功能及血清TNF-α,IL-1和IFN-γ含量均显著提高(P<0.01),并呈一定量效关系。结论AVPF-I能抑制H22肝癌生长,其抑瘤作用很可能是通过对荷瘤小鼠免疫功能的增强实现的。
Objective To study the anti-tumor effect of the proteoglycan extracted from abalone viscera (AVPF-I). Methods Hepatic carcinoma cell H22-bearing mice were randomized to negative control (physiological saline), positive control (cyclophosphamide) and three doses of AVPF-I groups. The tumor growth, cell mediated immune function including lymphocyte proliferation, phagocytosis of peritoneal macrophage, NK cell activity, and the level of serum TNF-α, IL-1 and IFN-γ were detected. Results A significant inhibition of the tumor growth was observed in the AVPF-I group (P〈0.01) , with an inhibitory rate of 62.37%. Compared to the negative control group, a significant increase in relative spleen weight, lymphocyte proliferation, NK cell and phagocytotic activity of peritoneal macrophage in the AVPF-I group were also observed (P〈0.01). Moreover, the level of TNF-α, IL-1 and IFN- γ in the serum of the AVPF-I group were also enhanced (P〈0.01). All the above results exhibited in a dose-dependent manner. Conclusion The polysaccharide extracted from abalone viscera AVPF-I had anti-tumor activity, which was probably mediated by immunomodulation actions.
